Lyell Immunopharma, Inc. is a clinical-stage cell therapy company developing chimeric antigen receptor (CAR) T-cell product candidates for hematologic malignancies and solid tumors. Its lead candidate, rondecabtagene autoleucel (ronde-cel), is an autologous dual-targeting CD19/CD20 CAR T-cell therapy under development in the pivotal PiNACLE trial in the 3L+ setting, a Phase 1/2 trial in the 2L setting, and a second pivotal trial, PiNACLE-H2H, a Phase 3 randomized controlled head-to-head trial for large B-cell lymphoma in the 2L setting. Lyell uses approaches such as c-Jun overexpression and NR4A3 gene knockout to confer resistance to exhaustion, Epi-R to generate stem-like T cells with reduced exhaustion and improved proliferation and antitumor activity, and CD62L positive enrichment to generate CAR T cells with enhanced antitumor activity.
