Acrivon Therapeutics will deliver an oral and a poster presentation at the AACR D3 Conference highlighting its AP3 platform and the clinical-stage WEE1/PKMYT1 inhibitor ACR-2316. The presentations show how AP3 identified WEE1 inhibition-induced PKMYT1 activation as a resistance mechanism and guided the design of ACR-2316 to achieve sustained CDK1, CDK2, and PLK1 activation for potent tumor cell death. Early clinical data from the ongoing Phase 1/2 trial of ACR-2316 have demonstrated partial responses in AP3-predicted tumor types including small cell lung cancer, squamous non-small cell lung cancer, and adenosarcoma NSCLC, which have not previously shown sensitivity to other WEE1 or PKMYT1 inhibitors. The compound has also shown a favorable tolerability profile with adverse events primarily limited to transient, mechanism-based neutropenia and a notable absence of non-hematological adverse events.
