Sichuan Kelun-Biotech Biopharmaceutical announced that translational research results on its TROP2-directed antibody drug conjugate sacituzumab tirumotecan combined with osimertinib as first-line treatment for advanced EGFR-mutant non-small cell lung cancer have been published in Cancer Cell. The study, led by Professor Li Zhang and Professor Wenfeng Fang from Sun Yat-sen University Cancer Center, shows that EGFR tyrosine kinase inhibitors upregulate TROP2 in drug-tolerant persister cells, and that sac-TMT plus osimertinib inhibited persister cell formation and delayed tumor recurrence in preclinical models. Building on these findings, the company is conducting a Phase III registrational study of sac-TMT plus osimertinib versus osimertinib monotherapy in first-line advanced EGFR-mutant non-squamous NSCLC, with enrollment in China completed and the study now in follow-up. A separate Phase II study is evaluating the same combination as neoadjuvant treatment for EGFR-mutant resectable NSCLC. Sac-TMT is already approved in China for advanced EGFR-mutant NSCLC after progression on TKI therapy or TKI therapy and platinum-based chemotherapy, and has demonstrated significant progression-free survival and overall survival benefits in TKI-resistant settings.
